Well Lee, here is the thing. I am assuming the machine already has an active connection to the net and you may or may not have Spybot already loaded (see Process List or Taskbar). Spybot can be tricky sometimes but the good news is, Spybot has multiple locations where you can download definitions from.
Go ahead and start the updater (Start>Programs>Spybot blahblah>Updater and it will launch and give you the screen to press "Display beta updates", normally I wouldn't suggest it but let's shake things up and can't really hurt so press the box to make it a check mark and press 'Search'.
Now if you get an error before getting to the screen where you can check/download from the Mirror's, you can try rebooting in to safe mode but most likely should re-install or 'Repair' the installation and make sure you have the latest version (not definitions but version) by checking safer-networking's website.
If you have an old version, sometimes things won't work properly or at all but if you can give me more information as to errors or what occurs now, happy to help.
